NET Formation Was Reduced via Exposure to Extremely Low-Frequency Pulsed Electromagnetic Fields

Fracture-healing is a highly complex and timely orchestrated process. Non-healing fractures are still major clinical problem treatment remains difficult. A 16 Hz extremely low-frequency pulsed electromagnetic field (ELF-PEMF) was identified as non-invasive adjunct therapy supporting bone-healing by inducing reactive oxygen species (ROS) Ca2+-influx. However, ROS Ca2+-influx may stimulate neutrophils, the first cells arriving at wounded site, to excessively form neutrophil extracellular traps (NETs), which negatively affects healing Thus, this study aimed evaluate effect of ELF-PEMF on NET formation. Neutrophils were isolated from healthy volunteers exposed different NET-stimuli ELF-PEMF. NETs quantified using Sytox Green Assay immunofluorescence, with fluorescence probes. In contrast mesenchymal cells, exposure did not induce in neutrophils. result basal or enhanced PMA-induced formation but reduce amount DNA released. Similarly, induced LPS H2O2 reduced through As release affect can be started immediately after fracture treatment.
